摘要:NOVELTY - Production of highly pure amylase involves pre-treating starch solution with alkaline solution, agitating, leaving, treating pretreated starch solution with hydrochloric acid, maintaining in water bath at 60-80 degrees C, gelatinizing, adjusting pH, adding pullulanase, heating, inactivating enzyme, separating purified amylase, lowering temperature of starch solution, adding butanol, precipitating amylase molecules, stirring, cooling, slowly stirring, removing supernatant, filtering, ethanol washing and drying. USE - Production of highly pure amylase (claimed). ADVANTAGE - The method enables production of highly pure amylase having purity of more than 90% by simple and economical process. DETAILED DESCRIPTION - Production of highly pure amylase involves pre-treating starch solution with 0.4-0.6 mol/l alkaline solution, agitating, leaving at 20-30 degrees C for 5-20 minutes, treating pretreated starch solution with solution of hydrochloric acid, where 4-6% hydrochloric acid solution is added to total mass of the starch solution, maintaining in water bath at 60-80 degrees C for 30-90 minutes, gelatinizing, adjusting pH to 4-5, adding 12.5-100 U/g pullulanase, heating above 85 degrees C, inactivating enzyme for 10-30 minutes, separating purified amylase, lowering temperature of starch solution to 65-75 degrees C, adding 40-70 ml/l n-butanol, precipitating amylase molecules, stirring for 45-75 minutes, cooling to 40 degrees C, slowly stirring for 18-24 hours, removing supernatant, filtering resulting precipitate by suction, performing ethanol washing 3-4 times, drying in blast oven at 50-70 degrees C.
