摘要:研究丝素蛋白作为动物细胞生长基质和生物材料对于细胞生长的促进作用,探索丝素薄膜性能的进一步优化。通过细胞计数和噻唑蓝(MTT)法实验表明,丝素薄膜上猪髋动脉内皮细胞生长的密度是普通细胞培养基质上的242.01%,细胞在薄膜上的贴壁率为在普通基质上的238%。经由酸性成纤维细胞生长因子(aFGF)处理后,薄膜上的细胞生长密度是未经处理薄膜上的115.98%,细胞的贴壁率为在普通细胞培养基质上的269%。因此,丝素蛋白薄膜可作为动物细胞生长基质,经aFGF处理的薄膜显示更优越的细胞亲和性。
The study of silk fibroin as a membrane substratum for cultivation of animal cells and the improvements of its performance was carried out by cell counting and 3-(4,5-Dimethyl-2-1hiagolyl)-2,5 diphenyl 2H · tetragolium bromide(MTT) assay method during the cell culture under different conditions when porcine iliac artery endothelial cell line (PIEC) was used as the model cell. Results show that the cell density on the membrane is 142.01% higher than that on the normal plate, and the cell density on the silk fibroin membrane treated with acidic fibroblast growth factor (aFGF) is 15.98% higher than that on this membrane untreated. The cell adhesion rates on the membrane and the membrane treated with aFGF are 138% and 169% higher than those on the normal plate respectively. It was proved that silk fibroin manifesteditself as an active substratum for cultivation of animal cells and the membrane treated with aFGF was better.
