摘要:目的:建立当归水溶性成分的HPLC指纹图谱方法。方法:采用梯度洗脱法,对当归水溶性成分进行HPLC测定,采用Phenomenex Luna C18(250 mm×4.6 mm ID,5μm)色谱柱,以水(0.1%甲酸)-乙腈作为流动相,检测波长254 nm,洗脱流速0.5 mL/min,记录时间65 min。结果:对指纹图谱方法学的精密度、重复性、稳定性考察中特征峰相对峰面积、相对保留时间的RSD均小于5%。本试验把HPLC指纹图谱结合相似度分析和聚类分析用于当归药材的质量评价,较好地表达了产地和用药部位信息。结论:本试验建立的HPLC指纹图谱方法可作为当归药材水溶性成分的质量控制手段。
AIM : To establish the HPLC fingerprint method of the water-soluble constituents in Angelica Sinensis. METHODS: A Phenomenex Luna C18 column (250 mm×4.6 mm ID, 5μm) was utilized. Water containing 0.1% formic acid and acetonitrile were used as gradient eluents. The detection wavelength was set at 254 nm and the flow rate was 0.5 mL/min. UV spectra was recorded within 65 min. RESULTS : The RSD of relative peak areas and retention time of peaks in precision, repeatability and stability has not reached 5.0%, respectively. HPLC fingerprint, combined with similarity analysis and cluster method, are used to evaluate the quality of Angelica Sinensis, and it describes the character of regions and fractions properly. CONCLUSION : The HPLC fingerprint method is practicable as a standard method in controlling the quality of the water-soluble constituents in Angelica Sinensis.
