摘要:NOVELTY - Serum-free culture medium comprises 21 amino acids e.g glycine and histidine, 6 vitamins e.g biotin and folic acid, 8 salts e.g magnesium chloride, magnesium sulfate and potassium chloride, 8 lipoids e.g cholesterol, myristic acid and palmitic acid, 4 microelements e.g ferric nitrate and green copperas, 2 buffers sodium bicarbonate and hydroxyethyl piperazine ethane sulfonic acid, 1 protein hydrolyzate e.g trypsin hydrolyzate, 1 acid-base indicator e.g phenol red and other additives e.g glucose, hypoxanthine and albumin. USE - The serum-free culture medium is useful for adherent growth of Madin-Darby canine kidney epithelial (MDCK) cell and single cell suspension growth of MDCK cell for producing useful biological products in industries. ADVANTAGE - The serum-free culture medium is specific, has improved quality and enables long period subculture of MDCK cell simply. DETAILED DESCRIPTION - Serum-free culture medium comprises 21 amino acids, 6 vitamins, 8 salts, 8 lipoids, 4 microelements, 2 buffers, 1 protein hydrolyzate, 1 acid-base indicator and 6 other additives. The amino acids are chosen from aminopropionic acid, arginine, asparagine, aspartic acid, cysteine, sulfo-aminolactic acid, glutamic acid, glutamine, glycine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, proline, serine, threonine, tryptophan, tyrosine and valine. The vitamins are chosen from biotin, folic acid, nicotinamide, pyridoxine, thiamine and lipoic acid. The salts are chosen from magnesium chloride, magnesium sulfate, calcium chloride, potassium chloride, sodium chloride, disodium hydrogen phosphate, sodium dihydrogen phosphate and sodium pyruvate. The lipoids are chosen from cholesterol, tocopheryl acetate, myristic acid, palmitic acid, palmitoleic acid, stearic acid, Tween 80 (RTM: Polysorbate 80) and segmented polyether F68. The microelements are chosen from copper sulfate, ferric nitrate, green copperas and sodium selenite. The buffers are chosen from sodium bicarbonate and hydroxyethyl piperazine ethane sulfonic acid. The protein hydrolyzate is the trypsin hydrolyzate and acid-base indicator is phenol red. The other additives are chosen from glucose, hypoxanthine, thymidine, transferrin, albumin and insulin.
