摘要:NOVELTY - Preparing dihydroxyacetone comprises performing modification of Gluconobacter oxydans, which oxidizes glycerol by performing biotransformation reaction. The modification of Gluconobacter oxydans is performed to increase sldAB gene expression, and knocking out mgdh gene and madh genes. USE - The method is useful for preparing dihydroxyacetone. ADVANTAGE - The method is economical as it utilizes only glucose as carbon source and it has high production efficiency. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is also included for transforming Gluconobacter oxydans for converting glycerol to dihydroxyacetone (preparing genetically engineered bacteria of Gluconobacter oxydans), comprising (i) utilizing gene targeting technology to construct a fixed point Gluconobacter oxydans, where the construct comprises two mgdh gene segments as homologous recombinant arms and at least one selective marker gene fragment, (ii) utilizing gene targeting technology to construct a fixed point Gluconobacter oxydans, where the construct comprises two madh gene segments as homologous recombinant arms and at least one selective marker gene fragment, (iii) increasing in the level of expression of sldAB gene in Gluconobacter oxydans, and (iv) utilizing adaptive evolution to obtain genetically engineered bacteria (mutants) by using only glucose as the sole carbon source in the culture medium.
