摘要:以金针菇(Flammulina velutipes)多糖保留率和蛋白质清除率为评价指标,采用正交试验优化壳聚糖絮凝工艺,絮凝最佳工艺条件为:提取液浓缩比3:1,絮凝剂用量2 mL,絮凝时间15 min,此工艺条件下得到多糖保留率和蛋白质清除率分别为86.90%和60.52%,多糖得率为水提醇沉工艺的2.7倍。
Orthogonal testing was used to optimize conditions for isolating Flammulina velutipes polysaccharide (FVP) from aqueous extracts by flocculation using chitosan. FVP material of highest purity (86.9%) was obtained after 15 minutes flocculation following the addition of 2 mL chitosan solution (1% w/v in 1% acetic acid solution) to 50 mL of concentrated (three-fold) aqueous extract. This represented a 2.7- fold increase compared with yields obtained using alcohol precipitation.
