摘要:多粘类芽孢杆菌HY96-2发酵液经乙醇沉淀,Sevag法去蛋白,透析和DEAE-52离子交换柱等手段纯化,获得HY96-2胞外多糖组分B和寡糖组分D,其中多糖组分B纯度为91.39%。进而采用凝胶渗透色谱法(GPC)、气相色谱法(GC)测定了多糖组分B的分子量,得组分B的分子量为1.22×10^6 Da。实验对组分B的单糖组成进行了分析,确定其含有5种单糖:鼠李糖、木糖、甘露糖、半乳糖、葡萄糖,它们的摩尔比为:2.52∶0.25∶1∶1.61∶2.24,为一种新结构多糖。
By ethanol precipitation,Sevag method to remove protein,dialysis and DEAE-52 anion exchange column chromatography,the extracellular polysaccharides of Paenibacillus polymyxa HY96-2 have been purified.Components B and oligose D were isolated and purified from the extracellular polysaccharides.The purity of component B is 91.39%.The molecular weight of component B is 1.22×10^6 Da,which was analyzed by gel permeation chromatography and gas chromatography.The monosaccharide composition of component B was analyzed.It contains 5 kinds of monosaccharides:rhamnose,xylose,mannose,galactose and glucose,and the molar ratio is 2.52∶0.25∶1∶1.61∶2.24.The structure of component B is new.
