摘要:通过COⅠ序列相似性分析、系统发育关系分析和形态分类方法对从阿根廷进口原木中截获的火蚁属昆虫进行种类鉴定。对截获的火蚁虫体DNA进行扩增和测序,获得火蚁的线粒体COⅠ基因序列,并与GenBank中相应的火蚁属序列进行比对分析。结果表明,截获火蚁线粒体COⅠ基因序列与GenBank中Solenopsis interrupta的序列相似性为92.96%~98.39%,而与红火蚁S.invicta(EU311556)、S.saevissima(AY950779)和热带火蚁S.geminata(AY254489)的序列相似性分别为93.84%、95.85%和87.18%。以NJ法构建的系统发育树显示截获火蚁与S.interrupta最为接近,两者位于同一亚组。根据以上分析及观察结果,将截获火蚁SHCIQFA1-2鉴定为Solenopsis interrupta。
The partial sequence of mitochondrial cytochrome oxidase subunit I( CO I) of fire ant ( SH- CIQFA1-2) intercepted in logs from Argentina in 2007, was amplified and sequenced in order to identify the species. The result of sequence analysis showed that the similarity of CO I gene sequence is 92.96% - 98.39% between SHCIQFA1-2 and S. interrupta, and sequence similarity of 95.85%, 93.84% and 87.18% with S. saevissima ( AY950779 ), S. invicta ( EU311556 ) and S. geminate ( AY254489 ), respectively. The phylogenetic relationship tree from CO I gene sequence revealed the fire ant (SHCIQFA1-2) and S. interrupta were in the same subgroup. The fire ant (SHCIQFA1-2) was identified as S. interrupta based on the morphological characteristics, sequence analysis and phylogenetic relationships.
