摘要:建立了冬虫夏草固体发酵粉中以麦角甾醇含量为生物量指标的分析方法:麦角甾醇含量的RP-HPLC检测方法.采用超声和回流等方法处理样品.结果:虫草粉的乙醇提取物中游离麦角甾醇的含量为53333±603μg/g,总麦角甾醇的含量138146±1896μg/g,HPLC分析采用麦角甾醇标准品外标定量法,线性范围:057~14317μg/mL,直线回归分析方程:Y=1879X-2E-13,R2=1.精密度相对偏差(RSD):0361%.样品中麦角甾醇的含量稳定,重现性好,可以作为冬虫夏草固体发酵的生物量的控制指标.
With reversed-phase high-performance liquid chromatography (RP-HPLC),a method to determine ergosterol in extract of Cordyceps Sinensis mycelium solid fermentation preparations had been developed.The fermentation preparations were extracted by ultrasonic or circumfluence.The dissociative ergosterol was 533.33±6.03?μg,and total ergosterol was 1?381.46±18.96?μg.The linear range of the HPLC method was 0.57~143.17?μg/mL.The regressed equation was Y=18.79X-2E^(-13),R^2=1.The precision RSD was 0.361%.The ergosterol was stable and replicated well in the samples.it could be used as a control parameters in the determining of biomass of the solid fermentation preparations.
