摘要:包涵体(inclusion body,IB)曾被认为缺乏正确的分子结构。近年来,傅里叶变换红外光谱(FT-IR)成为对包涵体蛋白质进行结构分析的有效手段,利用傅里叶变换红外光谱对包涵体蛋白质进行结构分析,表明IB蛋白质含有天然或类天然结构。所谓的包涵体蛋白质的品质,是指与复性率相关的IB蛋白质状态,主要反映为包涵体蛋白质构象的差异性。影响包涵体蛋白质品质的,既有内因也有外因,分析其影响因素,将有助于调整重组蛋白质生产的最佳培养条件。活性包涵体是近年来包涵体研究领域最具意义的成就之一。对包涵体蛋白质品质的研究将对包涵体蛋白质的复性以及活性包涵体的研究和应用具有重要意义。
Inclusion body (IB) is once being widely believed to be lack of correct molecular structure. In the past few years, it is indicated that inclusion body proteins contain native or native-like structure by FT-IR spectroscopy which is an effective method to analysis the structure of inclusion bodies. IB quality refers to a state correlated with its refolding yield, which are mainly regarded as conformational differences of IB protein. Analyzing the internal and external influence factors of the IB quality will help to regulate the optimized culture conditions to produce the recombinant proteins. Active IB is one of the most significant achievements in the area of IB study. Researches of IB quality play an important role on the renaturation of IB and the application of the bioactive IB.
