摘要:以生物相容性良好的亲水性高分子材料作为水凝胶的骨架材料,制备新型目标药物水凝胶贴剂;然后以亲水性高分子材料、填充剂、保湿剂、交联剂和交联调节剂等含量作为试验因素,以剥离强度和黏着力作为考核指标,采用均匀设计试验法优选制备目标药物水凝胶贴剂的最优方案。研究结果表明:以离体大鼠皮肤为屏障进行经皮渗透试验,用高效液相色谱(HPLC)法测定接收池中药物浓度,并计算药物累积释放率(Q),则Q和时间(t)之间呈良好的线性关系,符合释药过程中0级释放过程;该目标药物水凝胶贴剂具有良好的黏着力、剥离强度、载药性且对皮肤刺激性较小。
With hydrophilic polymer with good hiocompatihility as skeleton material of hydrogel,a new hydrogel patch for target medication was prepared. Then,with hydrophilic polymer,filler,humectant,cross linker and crosslinking regulator contents as experiment factors,peeling strength and adhesion as evaluation indexes,the optimal project of preparing hydrogel patch of target medication was preferred by unitform design experiment. The research resuhs showed that the relationship between Q(accumulaling release rate ot' drug) and t(time) was a good linear relationship,and it accord with zero level release process in release process of drug when percutaneous penetration experiment was implemented with isolated rat skin as harrier,the drug concentration in receiving pool was tested by high performance liquid chromatography (HPLC),and Q value was calculaled. The hydrogel patch of target medication had good adhesion ,peeling strength,drug loading and smaller skin irritation.
