摘要:NOVELTY - Papain is separated and purified by dissolving papain-containing raw material, removing insoluble substances, and adjusting concentration of protein to 10-20 mg/ml to obtain an initial crude enzyme solution; mixing initial crude enzyme solution with polyethylene glycol, inorganic salt and water and splitting phases when components are mixed up; and recovering papain from polyethylene glycol phase after phase splitting by ion exchange. The content of the initial crude enzyme solution is 30-40 mass%. The content of polyethylene glycol is 10-30 mass%. USE - Method for separating and purifying papain (claimed). ADVANTAGE - The method has high biocompatibility and simple and moderate operation. It is easy to expand in procedure, low in loss of enzyme activity in product, and free of remnant of organic solvent, and simplifies immobilizing steps. DETAILED DESCRIPTION - Papain is separated and purified by dissolving papain-containing raw material, removing insoluble substances, and adjusting concentration of protein to 10-20 mg/ml to obtain an initial crude enzyme solution; mixing initial crude enzyme solution with polyethylene glycol, inorganic salt and water and splitting phases when components are mixed up; and recovering papain from polyethylene glycol phase after phase splitting by ion exchange. The content of the initial crude enzyme solution is 30-40 mass%. The content of polyethylene glycol is 10-30 mass%. The content of inorganic salt is 10-25 mass%. The pH is 5.5-6.5 INDEPENDENT CLAIMS are included for: (1) the papain obtained by above method; and (2) method for immobilizing papain which comprises immobilizing papain in polyethylene glycol phase enriched with papain obtained after phase splitting method.
