摘要:羟丙甲纤维素(HPMC)凝胶点是影响亲水骨架缓释片缓控释效果的一个重要物理指标。HPMC凝胶点高,则水化速率快,越易形成凝胶层,控释效果越好。然而,现有的HPMC凝胶点测量方法主观误差很大。为解决这个问题,本室自主研制了凝胶点测量仪器,它可通过程序升温引起溶液黏度变化以确定HPMC凝胶点,为凝胶点的测定提供了一种精确的测量方法,也填补了市售黏度计不能用于程序升温的黏度测量的空白。用自制凝胶点测定仪测定了两种型号(60HD4000和75HD4000)HPMC的凝胶点,所得结果与标示值基本一致。并采用上述型号HPMC分别制备了茶碱缓释片。体外释放度试验结果表明,采用凝胶点较高的HMPC为骨架制得的缓释片缓释效果较好。
The gel point of hydroxypropyl methyl cellulose (HPMC) is an important physical index of influencing sustained-release effect in sustained-release hydrophilic matrix tablets. The hydration rate of the HPMC with higher temperature of gel point was faster, so that the formation of the gel layer was easier and the sustained-release effect was better. However, the existing measurement method of HPMC gel point had a significant subjective error. To solve this problem, a measuring instrument of gel point was independently developed to determine the gel point by changes in viscosity of the solution under programmed temperature. This instrument not only provides a precise measurement method to determine the gel point, but also fills the blank that viscosity could not be measured by commercial viscometer under programmed temperature. The gel points of two types of HPMC (60HD4000 and 75HD4000) were determined by the self-made instrument, and the obtained results were basically consistent to the labeled values. The theophylline sustained- release tablets were respectively prepared with above two types of HPMC. The results of in vitro release test showed that the sustained-release effect of the tablets with higher temperature of HPMC as the matrix was better.
