摘要:蛹虫草多糖具有抗氧化、抗肿瘤等多种药理活性,但其含量少、提取难度较大,因此研究高效、低成本的蛹虫草多糖提取工艺成为当前的研究重点和难点。本文将超声波和酶解两种技术进行结合,用于蛹虫草多糖的强化提取,以多糖得率为指标,先对提取过程中超声与酶解强化提取组合的方式进行优选,在此基础上通过单因素考察和响应面分析,优化组合工艺参数。结果表明:最优工艺条件为纤维素酶用量4.0%、超声功率180 W、提取温度50℃、提取液pH 4.5、液固比100 mL/g、提取时间60 min,该工艺条件下,蛹虫草多糖得率为22.00%±0.33%,比传统热水浸提法提高1倍以上。超声耦合酶解强化提取实验条件温和,在提高蛹虫草多糖得率的同时还缩短了提取时间,是一种高效、绿色的提取方法。
Cordyceps militaris polysaccharide is an important component in cordyceps militaris,which has a variety of pharmacological activities such as immunomodulation,anti-tumor,anti-virus,anti-infection.It is difficult to extract because of its low content.Therefore,the research on an efficient and low-cost extraction technology of Cordyceps militaris polysaccharide has become the current focus and difficulty.In this paper,ultrasonic and enzymatic hydrolysis enhanced technologies were combined to extract polysaccharide from Cordyceps militaris.Taking the yield of polysaccharides as the response value,the combination methods of ultrasonic and enzymatic hydrolysis in the extraction process were optimized first,and then the single factor investigation and the response surface analysis were used to optimize the process parameters.Results showed that the optimized extraction process of Cordyceps militaris polysaccharide was as follows:cellulase dosage of 4.0%,the ultrasonic power of 180 W,extraction temperature of 50℃,pH of 4.5,liquid-solid ratio of 100 mL/g,extraction time of 60 min,and the yield of Cordyceps militaris polysaccharide was 22.00%±0.33%,which was more than 1 time higher than that of the traditional water extraction method.Ultrasonic coupled enzymatic hydrolysis enhanced extraction method was an environmental and efficient method,which could greatly improve the yield of Cordyceps militaris polysaccharide and shorten the extraction time.
