摘要:研究了腺苷合成途径中不同前体氨基酸对芽孢杆菌ATCC21616产腺苷的影响。其中谷氨酸对腺苷合成的促进作用最大,能提高产苷30%以上。谷氨酰胺、丝氨酸、苏氨酸和甲硫氨酸也能促进腺苷合成,分别提高16%,14%,10%和6%。进一步研究添加谷氨酸后发酵液中NH4+浓度、有机酸浓度和胞内NAD+浓度的变化,发现添加谷氨酸能提供稳定的NH4+供应,并提供重要的代谢中间物α-酮戊二酸。同时,发酵液中乙酸和丙酮酸的浓度比对照低,胞内NAD+浓度较高,糖酵解生成的丙酮酸能顺利地进入TCA循环,从而能为菌体和腺苷合成提供充足的能量和前体,促进腺苷的合成。
Effect of different amino acid precursors of adenosine biosynthesis on adenosine production by Bacillus sp. ATCC 21616 was studied. Addtion of glutamate, glutamine, serine, threonine and methionine increased adenosine production by 30%, 16%, 14%, 10%, and 6% respectively. The cause of the promotion of adenosine production was further investigated. Results showed that glutamate addition could provide steady level of NH4^+ and a-ketoglutarate during fermentation. In addition, the concentration of acetate and pyruvate in the broth was lower, intracellular concentration of NAD+ was higher than that of the control, so the pyruvate synthesized from glycolysis could enter TCA cycle smoothly, the energy and precursor for cell growth and adenosine synthesis was enough.
