摘要:线粒体内过氧化氢(H_(2)O_(2))浓度异常与癌症、炎症和帕金森病等疾病密切相关,准确检测线粒体内H_(2)O_(2)水平意义重大。本文以1,8-萘酰亚胺为荧光团,苯硼酸酯为识别基团、季铵盐阳离子为线粒体靶向基团,设计合成了一种萘酰亚胺类荧光探针(CHN)用于对H_(2)O_(2)的检测以及细胞内活性氧的荧光成像。实验结果表明,CHN对H_(2)O_(2)具有较强的光谱响应,在1~100μmol/L范围内,体系的荧光强度与H_(2)O_(2)的浓度呈线性关系。方法对H_(2)O_(2)的检测限为0.84μmol/L(3σ/k);并可实现对细胞内源性和外源性H_(2)O_(2)的荧光成像,且具有良好的线粒体定位能力。
As an important reactive oxygen species,hydrogen peroxide(H 2O 2)primarily produced in mitochondria plays crucial roles in cell proliferation,differentiation and defense against pathogen invasion.Elevated H_(2)O_(2) levels are closely associated with some diseases such as cancer,inflammation and Parkinson’s.Therefore,it is highly needed to detect H_(2)O_(2) in mitochondria.Herein,a 1,8-naphthimide-based fluorescent probe(CHN)was designed for H_(2)O_(2) detection.Borate and quaternary ammonium cations were used as the recognition group and targeting group,respectively.The experimental results revealed that CHN exhibited significant spectral responses to H_(2)O_(2),with a detection limit of 0.84μmol/L(3σ/k).The probe could target in mitochondria and detect endogenous and exogenous hydrogen peroxide in living cells.
