摘要:从麦芽根中提取 5′-磷酸二酯酶 ,酶活可达 474U/ m L。将所得酶液用于水解酵母核糖核酸 ,得到 5′-CMP、UMP、GMP、AMP 4种单核苷酸。最佳反应条件为 :底物 RNA浓度为0 .0 1 g/ m L,每毫升底物的酶用量为 3 0 U,酶解温度 70°C,p H7.0 ,时间 2 h,5′-核苷酸转化率达80 %。
Extraction of 5′ phosphodiesterase from barley root was conducted. The enzyme activity obtained was 474 U/mL. When ribonucleic acids from yeast were digested with the enzyme solution, 4 kinds of nucleotides, 5′ CMP, UMP, GMP and AMP were resulted. The optimum conditions for the reactions were as follows: concentration of substrate RNA, 0.01g/mL; enzyme activity used for per mL of substrate, 30U; digestion temperature, 70°C; pH7.0; reaction time, 2h. The yield of 5′ nucleotide attained 80%.
