摘要:L-丙氨酸、过量草酸和乙醇在苯中同步进行乙酯化和草酰化制得N-乙氧草酰基-L-丙氨酸乙酯,再经环合、水解、脱羧制得4-甲基-5-乙氧基噁唑,进而与马来酸二乙酯经Diels-Alder加成、芳构化、还原后成盐得到维生素B6吡哆醇盐酸盐,总收率为56%。
Pyridoxine hydrochloride was synthesized from L-alanine, excess oxalic acid and ethanol in benzene by simultaneous esterification and acylation to afford ethyl N-ethoxyoxalyl-L-alaninate, which was subjected to cyclization, hydrolysis and decarboxylation to give 4-methyl-5-ethoxyoxazole, followed by Diels-Alder addition, aromatization, reduction and then HCl salt formation with an overall yield of 56 % (based on L-alanine).
