摘要:智能响应性纳米载体作为肿瘤治疗的新兴策略,因其高靶向性以及高生物相容性展现出显著的临床应用潜力。本文首先设计合成了两亲性共聚物:聚硫化丙烯-聚(3-丙烯酰胺基苯硼酸-co-聚乙二醇甲醚甲基丙烯酸酯)(PPSk-P(APBA_(m)-co-OEGMA_(n))),通过自组装形成具有活性氧(ROS)响应性的聚合物胶束;然后利用多酚类药物原花青素(PC)对亲水层P(APBA_(m)-co-OEGMA_(n))进行交联,形成硼酸酯共价交联网络,制得了微凝胶,从而得到具有pH/ROS双重响应性的微凝胶载体。通过动态光散射仪(DLS)、扫描电子显微镜(SEM)以及紫外-可见分光光度计(UV-Vis)等对微凝胶的粒径、表面形貌以及交联情况进行表征,并研究微凝胶对于疏水性小分子药物阿霉素(DOX)和亲水性大分子牛血清蛋白(BSA)的药物单载、多药共载及其药物释放动力学。研究结果表明,该pH/ROS双重响应性的微凝胶载体具有稳定的多药独立包载和良好的药物控释行为。
Smart responsive nanocarriers have emerged as a promising strategy for tumor therapy,demonstrating significant clinical potential due to their high targeting capability and excellent biocompatibility.An amphiphilic copolymer,poly(propylene sulfide)-poly(3-acrylamidophenylboronic acid-co-poly(ethylene glycol)methyl ether methacrylate)(PPSk-P(APBA_(m)-co-OEGMA_(n))),was designed and synthesized.Through self-assembly,reactive oxygen species(ROS)-responsive polymeric micelles were obtained.The hydrophilic P(APBA_(m)-co-OEGMA_(n))shell was further crosslinked using the polyphenolic drug proanthocyanidin(PC),forming a boronate ester covalent network that enhanced the stability of the nanocarrier while introducing pH responsiveness,and yielding dual pH/ROS-responsive microgel carriers.The particle size,surface morphology,and crosslinking behavior of the microgels were characterized using dynamic light scattering(DLS),scanning electron microscopy(SEM),and ultraviolet-visible(UV-Vis)spectroscopy.The drug-loading and release behavior of small-molecule doxorubicin(DOX)and macromolecular bovine serum albumin(BSA)were investigated,demonstrating efficient drug encapsulation and stimuli-responsive release.The results confirmed that the microgel carrier exhibited pH/ROS dual responsiveness,a stable independent loading of multiple drugs,and controlled drug release.This study provides a scientific foundation for precision drug delivery,intelligent release systems,and overcoming multidrug resistance in cancer therapy.
