摘要:为放大流感疫苗生产工艺至1500 L反应器规模,通过冷模实验研究了不同体积反应器条件下的混合时间、流体剪切力、体积氧传递系数和二氧化碳传递系数的区别。在1500 L反应器中进行培养时,转速为60 r/min,深层通气速率控制在22.5 L/min能够较好的满足供氧需求和提高CO_2的移除效果,同时也避免了剪切力对细胞的损伤。在1500 L反应器中病毒感染细胞60 h后,病毒滴度为2^(11)HA units/25μL,单位细胞病毒产量(Svy)为13145.05 virions/cell,2 L反应器的Svy为13298.70 virions/cell,两者仅相差1.16%。
In order to scale up the flu vaccine production to 1500 L animal cell culture bioreactor, cold form experiment was carried out in 1500 L bioreactor, including mixing time, liquid shear force, volumetric oxygen transfer coefficient and volumetric carbon dioxide transfer coefficient. When cultured in a 1500 L bioreactor, agitation speed and aeration rate was 60 r/min, and 22.5 L/min, respectively, which could meet the oxygen requirement, improve the effect of CO2 remove and reduce the shear force on cells. Virus-infected cells after 60 h in a 1500 L bioreactor, the virus titer was 211 HA units/25μL, the specific virus yield (Svy) was 13298.70 virions/cell, which is the difference of 1.16% in 2 L bioreactor
