摘要:以得率为指标,去离子水为溶剂,微波提取灰树花多糖,得到最佳提取工艺为:药材粒径840~2 000μm,液固比30 mL/g,微波提取210 s,溶剂pH=6;以脱蛋白率和多糖保留率为指标,壳聚糖为絮凝剂纯化灰树花多糖提取液,得到最佳工艺参数为:壳聚糖10 mL/g,絮凝1 h,液药比40 mL/g,药液pH=6,温度控制在45℃。与其他工艺相比,本文采用的微波提取及絮凝纯化方法操作高效,能有效提高灰树花多糖的得率和纯度。
Polysaccharides in Grifola frondosa were extracted by deionized water with microwave. Extraction yield as the index, optimum conditions were as follows:herbs diameter of 840-2 000μm, liquid-solid ratio of 30 mL/g, extraction time of 210 s, and solvent pH of 6.Polysaccharides were purified using chitosan as flocculant.With removal rate of protein and retention rate of polysaccharides as indexes, optimal parameters of flocculation purification were as follows:coagulant dosage of 1.0 mL/g, flocculation time of 1 h,liquid-drug ratio of 40 mL/g, drug solution pH of 6, and flocculation-temperature of 45℃. Compared with other operations, the method for microwave extraction and flocculation purification had the advantages of high operating efficiency, improving extraction yield and purity of polysaccharides.
