摘要:在酶存在下,用紫外光引发离子单体(甲基丙烯酰氧乙基三甲基氯化铵,METAC)聚合,一锅法制备了载酶聚电解质凝胶。利用红外、扫描电镜等表征了载酶凝胶的结构和组成,研究了温度和pH对固定化酶(Lipase@PMETAC)的催化活性影响。研究发现,在较宽的温度(25~50℃)范围和酸性条件下,固定化脂肪酶的催化活性均高于游离酶。酶促反应动力学结果表明,固定化酶的米氏常数(K_(m))更小,说明固定化酶与底物有更高的亲和力,证明聚电解质水凝胶为酶促反应提供了更适宜的反应环境。同时,凝胶的封装提高了酶的稳定性和循环使用性能。发展了一种聚电解质水凝胶固定化酶的新方法,实现了酶的稳定封装、催化活性保持和稳定性提升,为促进酶的催化应用奠定了基础。
This article involves the polymerization of ionic monomers(METAC)under UV light in the presence of enzymes,facilitating the one-pot synthesis of enzyme-loaded polyelectrolyte hydrogels.The structure and composition of these hydrogels were characterized using infrared spectroscopy and scanning electron microscopy.The study also examined how temperature and pH influence the catalytic activity of Lipase@PMETAC.The immobilized lipase demonstrates superior catalytic activity over the free enzyme across a broad temperature range(25~50℃)and under acidic conditions.Kinetic studies of enzyme reactions reveal a lower K_(m) for the Lipase@PMETAC,indicating a higher affinity between Lipase@PMETAC and substrate.This demonstrates that the polyelectrolyte hydrogel provides a more suitable reaction environment for enzyme-catalyzed reactions.Meanwhile,the encapsulation of gelenhances the stability and recyclability of the enzyme.This article developed a new method of immobilizing enzyme with polyelectrolyte hydrogel,which realized the stable encapsulation of enzyme,the maintenance of catalytic activity and the improvement of stability,and laid a foundation for promoting the catalytic application of enzyme.
