摘要:目的优化Marc145细胞在微载体上的生长条件。方法对微载体培养Marc145细胞的细胞接种密度、微载体浓度和细胞培养基等培养过程中的关键工程参数进行优化。结果以微载体细胞密度4×104个/mg、微载体浓度7mg/ml在高糖DMEM(含30mmol/L葡萄糖,6mmol/L Gln)培养基中培养,48h后每12h换液50%的培养方式效果最为理想。结论优化的培养工艺适用于病毒疫苗的生产。
Objective To optimize the condition for culture of Marc145 cells by microcarrier. Methods Analyze the influence of culture medium,density of cells inoculated and microcarrier concentration on the growth and metabolism of Marc145 cells. Results The optimal cell density and microcarrier concentration were 4 ×10^4 cells/mg microcarrier and 7 mg/ml respectively. The optimal culture medium was DMEM containing 30 mmol/L glucose and 6 mmol/L Gln. After the cells were cultured for 48 h,50% of the culture medium was changed every 12 h. Conclusion The optimal culture condition was suitable for the production of viral vaccines.
