摘要:研究了温度对大肠杆菌重组腺苷脱氨酶(ADA)的影响,发现37℃有利于ADA的表达,而30℃培养更有利于菌体生长并维持菌体活力。37℃菌体早衰可能与前期生长速度过快有关。通过对37℃条件下菌体前期生长进行控制,使后期的菌体生长达到了30℃培养时的生长趋势,表现出菌体活力强、细胞含水量高,对于大肠杆菌中重组ADA的表达起到了促进作用,蛋白表达水平提高了10%。
The effect of temperature on the production of adenosine deaminase (ADA) in E. coli was investigated. The results showed that 37 ℃ was benefit for the expression of ADA, while 30 ℃ was benefit for the growth of the cells. Through the growth control during the early cultivation stage at 37 ℃, the cell growth tendency reached the level of 30 ℃ cultivation. As a result, the expression level of ADA got promoted and the expressed quantity of adenosine deaminase was increased by 10%.
