ADVANTAGE - The fluorescent probe targets desired molecule with high precision, and has good applicability. DETAILED DESCRIPTION - Fluorescent probe comprises light-activated protein localization tag i.e. fluorescein derivative of formula (I).. An INDEPENDENT CLAIM is included for a method for preparing fluorescent probe, which involves: (A) reacting carboxyfluorescein with acetic anhydride for 2 hours; (B) purifying obtained product by column chromatography; (C) dissolving obtained product in tert.-butanol; (D) reacting obtained product with dimethylaminopyridine and di-tert.-butyl dicarbonate overnight at 40 degrees C; (E) drying obtained product in oven; (F) dissolving dried product in tetrahydrofuran; (G) adding nitro phenylchloroformate to obtain reaction product; (H) refluxing coumarin and selenium dioxide with para-xylene for 24 hours; (I) dissolving obtained product in methanol; (J) reducing obtained product with sodium borohydride; (K) purifying obtained product by column chromatography; (L) dissolving purified compound in mixture of water and methylene chloride; (M) adding nitrophenyl chloroformate to obtained product; (N) adding methyl-tert.-butoxycarbonyl-N'-ethylenediamine to obtained product; (O) purifying obtained product by column chromatography; (P) dissolving purified compound in purified compound in mixture of anhydrous dichloromethane and trifluoroacetic acid in volume ratio of 3:1 to obtain another reaction product; (Q) dissolving former and latter reaction products in anhydrous dichloromethane; (R) purifying obtained mixture by column chromatography using mixture of dichloromethane and trifluoroacetic acid in volume ratio of 1:1; (S) purifying obtained product by column chromatography; and (T) bonding obtained product with ligand to obtain the final product.
