摘要:利用表面活性剂与酶协同超声波法提取红藤中的总黄酮,以得率和抗氧化性为指标,考察表面活性剂的种类和用量、酶的种类和用量、提取温度和时间、溶剂pH、液固比等因素的影响;在单因素实验基础上运用正交试验确定最佳工艺。结果表明:以得率为指标的最佳工艺条件为提取时间70 min,液固比30 mL/g,纤维素酶用量0.5mg/mL,得率达到7.98%;以抗氧化性为指标的最佳工艺条件为提取时间70 min,液固比50 mL/g,纤维素酶用量0.5 mg/mL,此时自由基清除率为67.56%。
Ultrasonic extraction assisted with Tween-60 and cellulase was adopted to extract the flavonoids from Sargentodoxa.Factors influencing the extraction yield and the antioxidability property,such as the kind and the concentration of surfactant,the kind and the concentration of enzyme,extractive temperature and duration,solvent pH value,and the liquid to solid ratio were investigated.The optimal conditions were verified by the orthogonal experiment.The conditions for the optimal flavonoids extraction yield of 7.98% were extractive duration 70 min,liquid to solid ratio 30 mL/g,concentration of cellulase 0.5 mg/mL.As for antioxidability,the optimal conditions were extractive duration 70 min,liquid to solid ratio 50 mL/g,and concentration of cellulase 0.5 mg/mL,and the free radical scavenging capacity was 67.56%.
