摘要:在恒界面池中,从十六烷基三甲基溴化铵反胶束溶液中反萃牛血清白蛋白(BSA)的传质系数与搅拌转速无关,与反萃液的pH值和离子强度有一定关系,而温度的影响强烈(表观活化能达196kJ/mol)。表明该反萃过程属界面控制。提出了反萃BSA的界面过程机理,“满胶束”的聚结是速率控制步骤,而蛋白质与其周围带电的表面活性剂层的作用,对聚结速率有很大影响。
The overall masstransfer coefficients for reextraction of BSA out of CTAB-reversed micelles were measured using a stirred diffusion cell. They are unaffected by the stirring speed,associated with the pH value and ionic strength in stripping solution,and strong-ly influenced by the operative temperature with a formal activation energy 1 96 kJ/mol·These results showed that the reextraction is controlled by interfacial process.A mechanism of interfacial transport process in the reextraction of BSA is suggested.The coalescence of filled micelles with the phase interface is the rate-limiting step in the reextraction. However,the protein interaction with the surrounding charged surfactant layer has a very strong effect on the coalescence process.
