摘要:采用响应面分析法(RSM)对吐温60协同微波提取甘草黄酮的工艺进行优化。首先,考察6种非离子型表面活性剂(吐温20、吐温60、吐温80、司盘60、司盘80和聚乙二醇)对提取甘草黄酮的影响,最终选择吐温60为最佳活性剂。在单因素实验的基础上,通过Plackett-Burman(PB)实验设计筛选出对黄酮得率影响较大的3个因素为乙醇体积分数、提取时间和液固比,经响应面分析法对这些因素进一步优化。综合得率和利润两个要素,最佳工艺条件为乙醇体积分数30%、液固比35 mL/g、提取时间70 s、吐温60质量浓度0.01 g/mL和微波功率390 W。在此最优条件下,实测总黄酮得率为4.30%,与预测值4.26%相接近。
The technology of Tween 60 assisted microwave extraction of glycyrrhizic flavonoids was optimized by response surface methodology(RSM).The effects of six nonionic surfactants(Tween 20,Tween 60,Tween 80,Span 60,Span 80 and polyethylene glycol)on extraction of glycyrrhizic flavonoids were investigated,and Tween 60 was chosen as the best active agent.Then single factor test was performed,followed by Plackett-Burman(PB)design to select three main factors that had a greater impact on flavonoid yields(ethanol volume fraction,extraction time,liquid-solid ratio).These factors were further optimized by RSM.Considering profit and yields,the optimum extraction conditions were as follows:volume fraction of ethanol was 30%,liquid-solid ratio was 35 mL/g,extraction time was 70 s,mass concentration of Tween 60 was 0.01 g/mL,microwave power was 390 W.The yield of total flavonoids under the optimum conditions was 4.30%,which was close to the predicted value of 4.26%.
