摘要:他克莫司(1)作为一种广泛使用的免疫抑制剂,是由筑波链霉菌(Streptomyces tsukubaensis)发酵产生的。本试验通过在1生物合成过程中添加稀土元素,考察氯化镧、氯化铕和氯化铯对筑波链霉菌No.9993生物合成1的影响,并从添加氯化镧后其相关基因转录水平表达量的角度进行分析。结果显示,在发酵96 h添加10.0 mg/L氯化镧,1产量较对照提高37.5%,1生物合成过程中的结构基因fkb P的表达量显著增加,fkb O表达量也略有增加。提示氯化镧对1生物合成相关基因的转录有正向调控作用。
Tacrolimus (1), a widely used immunosuppressant, is produced by a fermentative strain Streptomyces tsukubaensis. Three rare earth elements, lanthanum (LaCl3), scandium (EuC13) and europium (CsCI), were added during the biosynthetic process to investigate their effects on the production of 1 by S. tsukubaensis No.9993, and the expression of the related genes after adding LaCl3 was analyzed. The results showed that the productivity of 1 increased 37.5 % compared with the control group when 10.0 mg/L LaCl3 were added at 96 h. The expression of structural genefkbP was remarkablely increased, while the expression of fkbO enhanced slightly. It suggested that lanthanum might play a positive role on the transcription of 1 structural gene.
