摘要:对芽孢杆菌的腺苷磷酸化酶基因deoD在大肠杆菌BL21(DE3)中的表达条件进行优化。确定在菌体培养的对数生长中期进行诱导,异丙基-β-D-硫代半乳糖苷(IPTG)诱导浓度为0.05mmol/L,诱导温度为37℃。腺苷磷酸化酶的最适pH为7.0,反应温度为50℃,其底物范围广,对腺苷、肌苷均有催化活性。
The expression of deoD gene from Bacillus sp. in E. coli BL21 (DE3)was studied. To achieve the high level of expression, IPTG was added in the middle exponential growth period up to 0.05mmol/L at 37℃. The properties of adenosine phosphorylase were investigated. The optimum pH value and temperature for this enzyme were 7.0 and 50℃. It was showed that this enzyme possessed a broad specificity to the substrates including both adenosine and inosine.
